IMAC Chromatography Resins

“IMAC Chromatography (Immobilized Metal Affinity Chromatography) is an extremely powerful technique for the purification of a specific class of target macromolecules, with the main advantage of this method being its high resolution power and separation efficiency. In the IMAC chromatography purification method, the target protein establishes a reversible interaction with a specific ligand attached to the chromatography support. Typically, metal immobilized on the support is used for the purification of recombinant proteins containing a polyhistidine sequence. Types of these supports include Ni-Agarose, NTA-Agarose, Co-Agarose, Fe-Agarose, Glutathione-Agarose, and p-Aminobenzamidine-Agarose. Supports containing glutathione through covalent attachment are utilized for the purification of GST fusion proteins. Since the GST portion of the fusion protein binds to glutathione with high affinity (unlike host proteins), this technique provides the basis for high purification efficiencies.”
