“In Ion-exchange Chromatography (IEX), proteins are separated based on differences in their surface charge. Purification relies on a reversible interaction between a charged protein and a support or ligand with an opposite charge (ionic interactions). In gel filtration chromatography, purified proteins become diluted, whereas in this method, they do not. The support (stationary phase) in this technique contains numerous acidic and basic functional groups. These basic ion exchangers consist of positively charged groups and are termed anion exchangers. Cationic exchangers carry negatively charged groups that attract positively charged molecules.
